Peptides for Brain Research

Understanding the brain is no easy task. This complex organ consists of more than a hundred billion neurons that are connected to one other in over a hundred trillion different ways. These connections are not static either, they are being formed and altered constantly. According to some estimates, every second of our lives these neurons are forming roughly a million new connections, called synapses[1]. The brain is unbelievably complex and while science is nowhere near to understanding it completely, a lot of progress has been made in recent decades toward understanding, treating, and even enhancing the brain.

A great deal of the progress toward understanding the brain can be attributed to peptides for brain research[2]. Peptides like Epithalon, Semax, Noopept, Cerebrolysin, Selank, FGL, Davunetide (NAP), Humanin and many others have helped scientists to probe the ways in which the brain grows, develops, fights off invading pathogens, and overcomes injury. These peptides have also helped us to understand how the brain controls appetite, regulates emotions, forms memories, and so much more.

One of the great revelations to come out of research into brain peptides is the understanding that many peptides carry out multiple functions in the central nervous system. In fact, the function by which most peptides were originally discovered is neither their most common nor their most important action. In other words, the role of peptides in the central nervous system is as complex as the structure of the central nervous system itself. There is a great deal still to be learned about the function and structure of the brain, which means peptides for brain research are becoming ever more popular in the scientific community.

What Do Peptides Do in the Brain?

Peptides are the primary components used in brain signaling cascades. They control both physiological and pathological phenomena including neurotransmission, neuron growth, neuromodulation, regulation of the neuroendocrine axis, regulation of blood flow in the brain, pain perception, mediation of the blood-brain barrier, and so much more. Though they are not a major component of the structure of the central nervous system, peptides basically control how that structure is created, maintained, and even destroyed[2].

Of the 80 or so natural peptides known to definitively affect brain function, more than 135 different effects have been elucidated. Such numbers barely scratch the surface though. Peptides do not simply work autonomously to alter brain function, but interact with other peptides and other components of the central nervous system. The interactions are complicated, but by using specific peptides for brain research, scientists are beginning to unravel some of the mysteries.

How Are Peptides for Brain Research Categorized?

In truth, it is nearly impossible to adequately categorize all the peptides for brain research. This is generally a result of the fact that most peptides have more than one function in the brain or a different function in the brain compared to other areas of the central nervous system or body. That said, it is useful to think about peptides for brain research based on their primary modes of action. Below are several categories of action that peptides for brain research might have along with some representative peptides for each group.

Peptides that Affect Learning and Memory

The largest category of peptides for brain research is clearly peptides that affect learning and memory. This is because common learning pathways can be stimulated in myriad different ways. Forming a memory can be triggered by several things from emotion to intentional efforts. For instance, peptides that affect sleep also affect learning, but that is not their direct function. To keep the category manageable, it is best to stick to peptides with direct effects on the rates of neuron growth and/or synapse formation as well as those that protect neuron health or modulate rates of synapse breakdown.

_Neuron Growth and/or Synapse Formation:_

In the first category are peptides like Semax and Selank. These peptides regulate gene expression in the brain and affect levels of brain-derived neurotropic factor (BDNF). BDNF is a primary regulator of the common pathway for neuron and synapse growth. Higher levels of BDNF are associated with greater long-term retention of information and activation of the default mode network (DMN). The DMN regulates concentration, memory, planning abilities, and our sense of self. Selank, in particular, has been shown to enhance memory stability and memory storage in mouse models[3], [4].

Epithalon also falls comfortably into this category. It is known to regulate gene transcription and, as a result, has been shown to increase rates of neurogenesis and neuron differentiation[5]. Epithalon has been shown to play important roles in the pineal gland, retina, and prefrontal cortex.

_Protection of Neuron Health:_

Selank and Semax can be thought of as regulating the breakdown of neurons because that is one of the roles of BDNF. However, other peptides have a more fundamental and important role to play in protecting synapses and neurons from degradation. Pinealon, for instance, acts as an antioxidant in the brain but also directly protects neurons from death following injury. Most importantly, it protects neurons from excitotoxic damage which occurs when levels of otherwise necessary neurotransmitters get too high. Humanin has similar function in protecting neurons from excitotoxic effects.

Noopept is another peptide known to protect the brain against excitotoxic injury following an excess of calcium release. Research shows that it can decrease the extent of necrotic damage that follows ischemia (e.g., stroke). Noopept, which easily crosses the blood-brain barrier because it is just two amino acids in length, has been shown to reduce memory deficits following brain injury, ameliorate the behavioral and cognitive consequences of dementia, and reduce anxiety[6], [7].

A new peptide in this category is Devunetide (NAP). Devunetide has been shown in animal studies to prevent apoptosis in neurons by stabilizing microtubule assemblies. This feature gives Devunetide neuroprotective, neurotrophic, and cognitive protective properties. It is of intense research focus because of its potential to treat conditions like Alzheimer’s disease, progressive supranuclear palsy, ALS, and more[8]. According to Professor Illana Gozes of Tel Aviv University, Davunetide has the capacity to both protect and restore microtubule function. Administration of Devunetide to Alzheimer’s disease patients has resulted in significant cognitive improvements. This makes Devunetide the first compound to cause any dramatic change in cognition in this disease and is opening up a whole new avenue for research into treating dementia[9].

The last peptide in this category is Cerebrolysin, which deserves special recognition because it has a multitude of different effects. Research shows that Cerebrolysin expands neuroplasticity, increases neuron survival, protects neurons from injury, and mediates the function of certain neurotransmitters. Cerebrolysin has all these effects because it isn’t a single peptide, but is rather an admixture of several peptides known to influence levels of BDNF, glial cell line-derived neurotrophic factor, ciliary neurotrophic factor, and nerve growth factor. Because of its wide-ranging properties, Cerebrolysin has been extensively researched in pain, stroke, dementia, and more.

Peptides that Affect Sleep

The quintessential peptide affecting sleep is delta-sleep-inducing peptide ( DSIP). Discovered in 1977, the original function identified for DSIP was the ability to alter sleep patterns. Since then, scientists have come to understand that DSIP can alter corticotropin levels, affect pain perception, lower stress levels, and normalize blood pressure. DSIP produces feelings of sleepiness, increases time spent sleeping by almost 60%, and shortens the time to sleep onset. New research is finding that DSIP may also have impacts outside of the central nervous system, affecting heart rate, the immune system, and energy balance.

Other peptides that affect sleep include Epithalon (Epitalon), Ipamorelin, Sermorelin, and Fragment 176-191. Epithalon, for instance, affects melatonin production and thus can have important influences on sleep onset, length of total sleep, and length of time spent in various stages of sleep. Ipamorelin, Sermorelin, and Fragment 176-191 all influence growth hormone signaling. Growth hormone, in turn, has been associated with orexin secretion. Orexin is a potent neurochemical that is fundamental in regulating sleep[10].

MK677, which is not a peptide but has action at the growth hormone secretagogue receptor, has been shown to increase stage 4 sleep duration as well as REM sleep duration. Because it acts to affect growth hormone levels, MK677 has many of the same effects on sleep that the above-mentioned peptides do.

One peptide that was not even thought to act in the central nervous system, but which is now understood to help synchronize light cues and regulate circadian rhythms, is Vasoactive Intestinal Peptide (VIP). VIP also helps to reduce inflammation in the central nervous system and regulate the accumulation of beta amyloid associated with Alzheimer’s disease.

Peptides that Affect Depression and Mood

Research shows that peptides like vasopressin, neuropeptide Y, and others affect the transmission of monoamines (e.g., serotonin, dopamine) in the central nervous system. These peptides are thought to be involved in several neuropsychiatric conditions and are often altered by common psychiatric treatments like electroconvulsive therapy (ECT) and SSRI medications. Interestingly, most of the knowledge about these peptides has come from research into other peptides that affect levels of neuropeptide Y, vasopressin, and the monoamines. These studies are revealing that mood is regulated by a complex set of chemical and structural interactions.

Many peptides not initially associated with depression and mood have, through research, been revealed to be useful in treating behavioral conditions. Ghrelin is one such peptide. It has been found to have a natural anti-depressant effect as a result of its effects on tryptophan metabolism in the brain. Tryptophan is a precursor to neurotransmitters like serotonin. Ghrelin analogues like ipamorelin, GHRP-2, and GHRP-6 are being tested to determine if their stimulation of the ghrelin receptor can precipitate similar changes in mood[11], [12].

Melanocyte Inhibiting Factor (MIF-1) may also be useful in the setting of the depression. This peptide, at low levels, has been shown to produce significant improvements in the symptoms of depression in a very short amount of time[13], [14]. The exact mechanism by which MIF-1 reduces symptoms of depression is not clear. It is thought that it acts as a positive allosteric modulator of several dopamine receptor subtypes and that it may also inhibit the release of other neuropeptides like alpha-MSH. This is interesting because sun exposure is associated with improved symptoms of depression, but has some effects on alpha-MSH that are opposite those of MIF-1. This partial contrast may make it easier for scientists to tease out the subtle neurotransmitter patterns that lead to depression so that they can develop more effective treatments.

Peptides for Brain Research: Summary

This article could go on into some detail discussing the multitude of peptides that affect the brain. Rather than go down that rabbit hole, however, it is instead targeted at a high-level overview of some of the peptides for brain research that are of most interest right now. Despite the ongoing work in this area, there is a great deal that needs to be done and much to learn about how peptides interact with the complex signaling of the brain to help improve cognition, reduce the effects of injury, alter sleep, change mood, and so much more.
