TB 500 and Hair Growth
The use of peptides to combat hair loss has gained traction in recent years and several have been studied for their potential to promote hair growth. These peptides work via several mechanisms, including stimulating the growth of hair follicles, altering hormone signaling, improving blood circulation to the scalp, and reducing inflammation. Among the peptides that are of interest to researchers investigating hair loss is TB 500. This peptide has generated some buzz in the scientific and medical communities due to its functional similarity to Thymosin Beta-4 and the fact that it is both easy and more affordable to produce. While initially investigated for its ability to promote hair growth, its applications in wound healing, injury recovery, and tissue regeneration have taken center stage. Recently, however, there has been a rise in research looking at the connection between wound healing and hair growth. This has led to renewed efforts to characterize TB 500 and its function.
Peptides like TB 500 offer promising possibilities for addressing various medical needs, from chronic wounds to musculoskeletal disorders and neurodegenerative diseases. The abilities of TB 500 to accelerate tissue healing, enhance wound closure, promote muscle repair, and support regeneration of neurological tissue in animal models are noteworthy. As a potent anti-inflammatory, TB 500 has a number of potential uses and thus a great deal of research has gone into understanding what it does and how it does it. Here is a look at how TB 500 is of interest to researchers attempting to understand hair growth and loss
What Causes Hair Loss?
Alopecia (another term for hair loss) can result from a variety of factors depending on the individual. In some cases, multiple factors may contribute to hair loss simultaneously. The most common cause of hair loss, in both men and women, is due to genetic factors that predispose a person to thinning hair. This is also known as androgenetic alopecia or male-pattern baldness. It tends to run in families and is characterized by a gradual thinning of hair usually on the crown of the head and at the hairline. Researchers know that this type of hair loss is caused by an interaction between the hormone dihydrotestosterone (DHT) and hair follicles that are genetically susceptible to its effects. DHT causes hair follicles to miniaturize over time, leading to shorter, finer, and less pigmented hair. Eventually, affected hair follicles may stop producing hair altogether. Interestingly, the propensity toward male-pattern baldness appears to be more closely related to a family history of baldness on the mother’s side.
Genetics are not the only cause of hair loss, however. Hormonal changes, medications, stress, nutritional deficiencies, and weight loss can all lead to hair loss. In the latter case, crash diets can lead to hair loss by inducing stress and causing nutritional deficiencies[1]. Addressing each of these conditions will often result in cessation of hair loss, but may _not_ lead to hair regrowth. To encourage hair to regrow, additional steps need to be undertaken. To this point, science has only had relatively ineffective topical and oral medications to offer those suffering from hair loss. A better understanding of the hair follicle, however, is helping to change that.
An Overview of the Hair Follicle
Scientists have revised their thinking. Rather than imagine the hair follicle as a simple structure, they now think of the it as a miniature organ of the skin, complete with its own complex hormonal regulation. The hair follicle is, after all, made up of nine different skin layers and at least five different cell types. Research in recent years has shown that hair growth in the follicle is only possible when the number of a certain type of cells, called dermal papilla cells (DPCs), reach a certain threshold in their development. By enhancing the number and differentiation of these cells, hair growth can be promoted. As it turns out, the role of DPCs in hair growth is very similar to how similar cells function in the process of wound healing. Scientists now understand that hair growth and wound healing share similarities in that they both require precise coordination among various cellular processes. These processes include cell proliferation (division and multiplication of cells), cell differentiation (specialization of cells into specific types), and cell migration (movement of cells to specific locations).
The transformation of hair follicles from the telogen (resting) stage to the anagen (growth) stage is dependent on interactions between DPCs and hair follicle (HF) stem cells. These interactions involve the secretion of signaling molecules by DPCs. This mimics the process by which certain cells of the immune system secrete signaling molecules that encourage endothelial progenitor cells to proliferate and differentiate. In both cases, signaling molecules (e.g. Notch, bone morphogenetic protein, sonic hedgehog) communicate with epithelial cells (the cells that line blood vessels) to encourage the growth of small blood vessels called capillaries. This enhanced nutritional supply then facilitates the transition of hair follicles from telogen to anagen phase. The same growth factors are associated with wound healing and their expression patterns have been shown to be enhanced via thymosin beta-4 administration.
The above explanation of the hair follicle is somewhat esoteric, but the general point is that the hair growth cycle is a complex process controlled by hormone and signaling molecule fluctuations in a complex organ. The similarities between the processes of wound healing and hair growth are providing scientists with a model for better understanding the latter. In particular, this model helps to make it clear why TB 500 has been so effective in the regulation of hair growth[2].
What Are the Available Options for Hair Growth?
Options for slowing hair loss or encouraging hair growth have grown steadily, particularly in recent years, as scientists work to understand the precise mechanisms that control the hair growth cycle[3]. The options that have been around the longest are the medications minoxidil and finasteride. Minoxidil is a topical medication while finasteride is taken by mouth in pill form.
Minoxidil works by encouraging blood flow to the scalp (improving nutrition to the hair follicle), prolonging the growth phase of hair, opening potassium channels, and increasing hair follicle size. Though the medication is generally considered to be well tolerated, minoxidil does have side effects and is far more effective in preventing hair loss than it is at encouraging regrowth following substantial loss. The use of minoxidil is a lifelong commitment as well because stopping the medication results in resumption of hair loss.
Finasteride is a 5-alpha-reductase inhibitor and prevents the conversion of testosterone into the hormone DHT. By lowering DHT levels, finasteride slows or stops the process of miniaturization that is connected to hair loss. Unfortunately, this highly effective medication comes with some rather unpleasant side effects including decreased libido and erectile dysfunction.
Non-medication options for hair growth have become more prevalent in the last decade. Low-level laser therapy (LLLT), hair transplants, and platelet-rich plasma (PRP) therapy have all been used with varying degrees of success to promote hair growth.
LLLT works on several levels to promote hair growth. Like minoxidil, it is thought to increase blood circulation to the scalp and promote nutrient delivery to hair follicles as a result. Research also suggests that LLLT may stimulate hair follicle growth by increasing energy production in cells, reducing inflammation, and mitigating oxidative stress. There is even some evidence to indicate that LLLT can influence DHT levels. LLLT is most effective in the early stages of hair loss, however, working more to mitigate loss than to encourage regrowth. Like minoxidil, LLLT treatment must be continued for the long term to remain effective[4], [5].
Hair transplants are the currently most effective way to mitigate hair loss. With the improvement of surgical techniques, they are a far cry from the much ridiculed “hair plugs” of the past. In the hands of a skilled surgeon, hair transplants can produce a natural-looking result that blends seamlessly with surrounding hair. This is also the only option to provide a permanent solution to male-pattern baldness because it utilizes hair that is resistant to the genetic effects that underly the condition. Transplant surgery works as well in the later stages of hair loss as it does in the early stages, which is another factor that sets it apart from the alternatives. Hair transplants are costly, however, and come with their own drawbacks such as the need for future transplants to replace hair that continues to be susceptible to hormone effects and the possibility of scarring.
Finally, platelet-rich plasma therapy is probably the newest option for promoting hair growth. It works best in the early stages of hair loss and must be performed periodically to maintain the benefits. It is most effective when used in conjunction with additional treatments, like minoxidil, and is relatively expensive. It has to be administered in a clinical setting due to the equipment required to separate platelets from other blood components[6].
Hair Growth Research
The options for encouraging hair growth have been limited until recently. And one of the reasons that TB 500 for hair growth is a topic of serious research interest is that existing treatments are expensive, time-consuming, and come with a variety of side effects. Current options help, but they are not the final solution. Fortunately, researchers have not been resting on their laurels. In recent years, there has been a strong interest in the ability of certain peptides to promote hair growth. Peptides that have been investigated include:
- GHK-Cu (Gly-His-Lys Copper Peptide): This peptide has shown promise in promoting hair growth and improving the overall health of the scalp. It may help increase the expression of growth factors and reduce inflammation in the scalp. GHK-Cu is a member of a larger group of copper peptides that have been shown to reduce inflammation and encourage cell division. Copper peptides, such as copper tripeptide-1, have also been studied for their potential to stimulate hair growth. They may work by increasing the size of hair follicles and promoting the proliferation of dermal papilla cells, which are important for hair follicle health[7].
- Biotinyl-GHK (Biotinyl Tripeptide-1): This peptide is a modified form of GHK-Cu and is believed to promote hair growth by improving hair follicle health and stimulating the anagen (growth) phase of the hair cycle. Biotinyl-GHK is found in a number of cosmetic and hair care products and is a derivative of GHK-Cu. Unlike GHK-Cu, Biotinyl-GHK does not chelate a copper atom. It does, however, contain a vitamin B additive (the biotinyl component) that is known to be beneficial to hair and skin health. It is often found in shampoo.
- Palmitoyl Pentapeptide-17: This peptide is sometimes found in hair growth serums and is believed to support the health of hair follicles and promote hair growth. It stimulates collagen production. Collagen is a component of healthy hair.
- Acetyl Tetrapeptide-3: This peptide has been studied for its potential to increase hair density and thickness by promoting the anagen phase of the hair growth cycle. Acetyl Tetrapeptide-3 is found in shampoos, conditioners, and other hair products because it is thought to reduce levels of DHT in the scalp. Research shows that it can produce stronger, more resilient hair by extending the growth phase. As with many of the hair growth options discussed, it works best when started early in the hair loss timeline and must be used continuously in order to maintain its effectiveness. It is often combined with other ingredients, including peptides on this list, to boost its effectiveness[8].
- Hexapeptide-11: This peptide has been investigated for its ability to stimulate hair growth by increasing the expression of growth factors and promoting the proliferation of hair follicle cells. Hexapeptide-11 is also a stimulator of collagen production with research suggesting that it improves both hair and skin health when used consistently. It is relatively inexpensive to produce via recombinant DNA technology that utilizes yeast.
TB 500 for Hair Growth
TB 500 is a derivative of thymosin beta-4, which is found naturally in most animals. In fact, TB 500 is the shortest version of thymosin beta-4 that retains the active domain of the naturally occurring peptide. It works by controlling actin polymerization, a critical component of cell mobility and cell structural integrity. Research shows that TB 500 is an important regulator of cell migration, particularly for cells of the immune and vascular systems. Research now shows that TB 500 plays a significant role in promoting hair growth by influencing critical events in the hair cycle. These include promoting the migration of stem cells and their progeny to the base of the follicle, facilitating stem cell differentiation, modulating the extracellular matrix, and encouraging blood vessel growth. Research in mice has revealed several promising benefits of TB 500 for hair growth.
Perhaps the most important role for TB 500 in promoting hair growth is its ability to stimulate the growth of blood vessels. This is particularly true of the small blood vessels, called capillaries, that supply oxygen and nutrients to hair follicles. VEGF (vascular endothelial growth factor) is the primary peptide controlling the development of blood vessels. It is known to be important in processes such as hair development, angiogenesis (the formation of new blood vessels), and recovery from cardiac injury. Tβ4, and by extension TB 500, induces VEGF expression[9], [10].
It isn’t just the expression of VEGF, however, that makes TB 500 a potent vascular growth stimulator. Research shows that it also activates vascular tubular formation, helping endothelial cells to grow, divide, and migrate to the locations in which they are needed. It also regulates another growth factor, called hepatocyte growth factor (HGF) that has been shown, despite its name, to influence the growth and differentiation of endothelial cells as well.
The ability of TB 500 to induce vascular growth is just one of the potential roles for the molecule in hair growth, however. As it turns out, the peptide has additional benefits such as promoting the migration and differentiation of HF stem cells, which are essential for the normal development of hair follicles.
There is also an abundance of evidence to indicate that TB 500 can regulate several signaling pathways to promote the synthesis of extracellular matrix components like matrix metalloproteinase-2 (MMP-2). This can lead to remodeling of the basement membrane in hair follicles, which allows the hair follicles to make better contact with the layers of the skin that contain blood vessels that deliver nutrients, growth factors, and oxygen.
Another signaling pathway that TB 500 has been shown to regulate is the Wnt signaling pathway. Wnt is the primary controlling pathway of hair follicle growth. Research indicates that TB 500 increases Wnt signaling to cause an increase in several other growth factors that regulate not only blood vessel growth, but hair follicle growth as well[2].
TB 500 for Hair Follicle Cell Survival
To this point, most of the attention surrounding TB 500 has focused on its ability to promote growth, primarily via cell migration and the enhancement of certain growth factors. A less discussed benefit of TB 500, however, is its ability to promote cell survival. Apoptosis, or programmed cell death, is a crucial process for maintaining tissue homeostasis but can become dysregulated in certain settings. Tβ4 has been shown to have anti-apoptotic properties, meaning it can help prevent or reduce cell death. This property is particularly important in the context of tissue repair and regeneration, where the survival of cells is critical for healing. As a process with a number of similarities to tissue regeneration, it should come as no surprise that hair growth is benefited by a reduction in apoptosis as well. In particular, TB 500 may help to offset the miniaturization process seen in hair follicles that are hypersensitive to DHT. This may, in turn, help to thwart the death of these hair follicles and interrupt hair loss.
The anti-inflammatory benefits of TB 500 factor heavily into its ability to regulate cell survival. It is well known that inflammation and oxidation are detrimental to cell survival and that both processes can be the result of dysregulated immune processes. Research has repeatedly supported the ability of TB 500 to not only suppress the production of pro-inflammatory cytokines like TNF-α and interleukin-6, but to increase the release of anti-inflammatory cytokines like interleukin-10. Research shows that TB 500 has a balancing effect on immune responses, helping to regulate process that, while beneficial, can sometimes become supercharged and do damage[11]–[13].
TB 500 Synergy for Hair Growth
Though research into the use of TB 500 in conjunction with other peptides and treatment options is limited, it is likely that future studies will reveal synergistic effects. In particular, TB 500 may make it possible to use smaller doses of currently effective medications, like finasteride, to achieve the same effects. This would lead to a reduction in the side effects associated with these medications and make them vastly more useful. Though TB 500 has not been directly explored in use with hair loss treatments, it has been explored as an adjuvant to antibiotic therapy.
Research shows that TB 500 can augment the effects of commonly used antibiotics like ciprofloxacin to improve outcomes in difficult to treat infections. TB 500 is thought to increase tissue penetration of antibiotics and reduce problematic inflammation that interferes with their actions. These mechanisms of action are highly applicable to hair growth as well. By increasing vascular permeation around hair follicles, for instance, TB 500 would make it easier for orally and topically administered hair growth treatments to reach their target areas. This would increase the efficacy of these treatments by getting more of the active ingredients to the sites where they are meant to have an effect.
By reducing inflammation, TB 500 would likely augment the benefits of treatments like LLLT and platelet-rich-plasma therapy. In the latter case, TB 500 would not only be synergistic with PRP in reducing inflammation, but would be pulling double duty by helping the injected PRP more effectively reach target areas to reduce inflammation and supply nutrients to hair follicles.
Finally, TB 500 could likely be used in conjunction with some or all of the peptides mentioned earlier. In particular, peptides like GHK-Cu and Acetyl Tetrapeptide-3. In the case of Acetyl Tetrapeptide-3 and TB 500, both are thought to help prevent miniaturization of the hair follicle, strengthen hair follicles, and thwart the actions of DHT. Both extend the anagen or growth phase of the hair cycle, and thus it stands to reason that if they work via different mechanisms then they could build upon the effects of one another to further extend anagen. Of course, the research in much of these areas remains to be done.
TB 500 and Hair Growth | Summary
TB 500 has long been known as a potent stimulator of wound healing and has been extensively studied for its ability to promote the growth, differentiation, and migration of cells that are important in regeneration. With all of the similarities between hair growth and wound healing, it should come as no surprise that research supports a role for TB 500 in stimulating hair growth and helping to prevent hair loss. TB 500 helps to reduce inflammation, improve nutrient delivery to hair follicles, stimulate hair follicle stem cell growth, and alter signaling pathways. Combined, these effects seem to have a prominent effect on hair growth as seen in animal studies. TB 500 needs to be investigated as both a stand-alone hair growth stimulator and as an adjuvant to existing hair loss treatments.
